ABSTRACT

The aim of this study was to evaluate the addition of vitamin C, reduced glutathione and the association thereof to the bovine semen cryopreservation extender. The ejaculate from nine bulls were divided into four fractions, each corresponding to a treatment, namely: control group-semen diluted with Tris-yolk extender; vitamin C group-semen diluted in Tris-yolk extender supplemented with vitamin C (2.5mmol/mL); glutathione group-semen diluted in Tris-yolk extender supplemented with reduced glutathione (2.5mmol/mL) and associated group-semen diluted in Tris-yolk extender supplemented with vitamin C (1.25mmol/mL) and reduced glutathione (1.25mmol/mL). Afterwards, the semen was packed into French straws and submitted to cryopreservation using automated equipment. After cryopreservation, the semen was thawed and evaluated considering sperm motility, morphology, plasma membrane, acrosome, mitochondrial potential and oxidative stress, as well as the thermo resistance test. Extender's supplementation with the association of vitamin C and reduced glutathione showed benefic effects on sperm motility and preservation of plasma and acrosomal membranes during semen cryopreservation, being also the group that showed higher values of reactive oxygen species. Thus, the association of both antioxidants contributed to the preservation of sperm cells in every analyzed characteristic, suggesting its use on bovine semen cryopreservation.(AU)

ABSTRACT

Determining sex may be more difficult in cases such as natural disasters, accidents or situations in which bodies are subjected to high temperatures, when individuals must be identified from their remains. The mandible is a very strong bone, presents high sexual dimorphism and may be useful in forensic identification. The object of the present study was to determine sex by metrical analysis of macerated mandibles of Brazilian adults. We analysed 113 fully dentate macerated mandibles of Brazilian adults, 47 belonging to women and 66 to men. We took 8 measurements using a digital calliper: bicondilar breadth (BC), bigonial breadth (BG), bimental foramina breadth (BM), distance between mental foramen and mandibular base (MF-MB), mandibular ramus height (MRH), maximum mandibular ramus breadth (MaRB), minimum mandibular ramus breadth (MiRB) and mandibular body length (MBL). The t test was used for statistical analysis of independent samples, and a ROC curve was constructed. Direct and stepwise discriminant analysis was carried out. SPSS v.22 software was used, with a significance threshold of 5 %. We observed that all the measurements presented statistically significant differences between the sexes, with greater mean values for men than for women. BG was the measurement which presented the greatest area under curve (AUC), and the highest correct prediction, followed by MRH and BC. The BM distance presented the smallest AUC and lowest correct prediction. The mean correct prediction was 85 % for direct discriminant analysis and 83.2 % for stepwise discriminant analysis, using the BG and MRH measurements. The measurements analysed in this study can be used to determine the sex of Brazilian individuals.

ABSTRACT

Bredemeyera floribunda roots are popularly used to treat snakebites in the semiarid region of Northeast Brazil, and previous studies indicate the anti-ophidian actions of triterpenoid saponins found in its roots. To assess B. floribunda root extract (BFRE) activity against the effects of Bothrops jararacussu venom (BjuV), antiphospholipasic, antiproteolytic, antihemorrhagic, antinecrotic, and anti-edematogenic activities were investigated in mice. Phytochemical analysis revealed the presence of saponins, flavonoids, and sugars, with rutin and saccharose being the major constituents of BFRE. Acute toxicity was determined and BFRE was nontoxic to mice. Phospholipase A2 and proteolytic activities induced by BjuV were inhibited in vitro by BFRE at all concentrations tested herein. BFRE (150 mg/kg) inhibited paw edema induced by BjuV (50 µg/animal), reducing total edema calculated by area under the curve, but carrageenan-induced paw edema was unchanged. Hemorrhagic and necrotizing actions of BjuV (50 µg/animal) were considerably decreased by BFRE treatment. Thus, BFRE blocked the toxic actions of B. jararacussu venom despite having no anti-inflammatory activity, which points to a direct inhibition of venom's toxins, as demonstrated in the in vitro assays. The larger amounts of rutin found in BFRE may play a role in this inhibition, since 3′,4′-OH flavonoids are known inhibitors of phospholipases A2.

ABSTRACT

The aim of this study was to evaluate hCG treatment on ovarian response and on pregnancy rate using a 9-day oestrus synchronization protocol in Santa Ines ewes. On a random oestrus cycle day, ewes received an intravaginal progesterone device (Primer-PR®, Tecnopec, Brazil). Nine days later (Day 9), 30µg of d-cloprostenol (Prolise®, Syntex, Argentina) and 250IU of eCG (Folligon®, Intervet, Brazil) were administered and the progesterone device was removed. This moment, the ewes were randomly assigned on two groups: Control Group and hCG Group. In the hCG Group, the ewes received 500IU of hCG (Vetecor®, Hertape-Calier, Spain) 24h after device removal. In the Control Group, the ewes did not receive any ovulation inductor. Control and hCG Groups ewes were inseminated 60h and 48h after device removal, respectively. There was no difference between the groups regarding the first ovulatory follicle diameter and the second ovulatory follicle. hCG Group ewes had shorter interval between device removal and ovulation (Control: 79.9±15.4h and hCG: 54.7±4.9h; P=0.001) and more synchronized ovulations. However, the treatment with hCG decreased the pregnancy rate after TAI (P=0,009). In conclusion, hCG administration improves ovulatory synchronisation, but causes a decrease in the pregnancy rate.(AU)

ABSTRACT

ABSTRACT

ABSTRACT

ABSTRACT: Dollarweed is a plant that is widely used by the Ayurvedic medicine and in the cosmetics industry because of its rejuvenating and revitalizing properties related to the nervous system and the brain. In order to evaluate the agronomic characteristics and the content of total flavonoids of the dollarweed, in response to the application of different levels of irrigation and shade, an experiment was conducted in an area at the coordinates 17º 47' 53'' latitude (S), 51º 55' 53'' longitude (W), with a mean altitude of 648m, from July to November. The climate is Aw (tropical), gently undulating relief (8% slope) with distroferric red latosol and clayey texture. The average annual temperatures are 18.1 ºC and 30 ºC. The average relative humidity of the air is 64.7% and the average annual rainfall is 1550 mm, with well-defined dry and rainy seasons. The assay was in randomized blocks, with a 2 x 2 factorial design, with 5 repetitions, amounting to twenty experimental units with two irrigation managements, being one with a daily application of water and the other with two daily applications of water. The shading treatments were under full sun and 50% shade. The fresh and dry weights of the aerial parts, the height and number of plants and the total content of flavonoids of the aerial parts were evaluated. The production of flavonoids was favored by the shade; irrigation did not influence the production of flavonoids or the agronomic characters evaluated.

ABSTRACT

The importance of the condyle-fossa relationship in the temporomandibular joint (TMJ) is not yet fully clear, however, some authors associate the non-central position of the head of the mandible in the mandibular fossa with temporomandibular disorder (TMD). Furthermore, other authors suggest that assessment of joint space is a diagnostic tool for assessing the TMJ alterations. The aim of this study was to evaluate the TMJ condylar position and joint space in Chilean individuals with TMD using cone-beam computed tomography. We selected 19 patients of "Unidad de Trastornos Cráneo Cervico Mandibulares (UCRACEM) - Universidad de Talca, Chile", who had a diagnosis of TMD according to the Research Diagnostic Criteria for Temporomandibular Disorders. The assessment of condylar position was performed using Pullinger & Hollender technique and to evaluate the anterior joint space (AJS), superior joint space (SJS) and posterior joint space (PJS) was used Ikeda & Kawamura technique. Posterior condylar position was found in 39.47% of condyles, central condylar position in 34.21% and anterior condylar position in 26.31%. To condyles in central position we found: AJS = 1.57 mm; SJS = 2.20 mm; PJS = 1.54 mm. To condyles in posterior position we found: AJS = 2.18 mm; SJS = 2.02 mm; PJS = 0.98 mm. To condyles in anterior position we found: AJS = 1.00 mm; SJS = 1.89 mm and PJS = 2.07 mm. We conclude that individuals diagnosed with TMD present anatomical regions predisposing to joint disorders with joint spaces diminished when compared with asymptomatic subjects. Furthermore, the posterior condylar position was the most common in individuals with TMD, however there was great variability in condylar position, therefore we suggest that a non-centralized position of the condyle is not necessarily associated with the presence of TMD. We further conclude that in individuals with TMD the condyles located in central and posterior position present EAP decreased compared with healthy individuals.

ABSTRACT

Therapy by low-level laser (LLL) or ultrasound (US) are commonly used as treatment after nerve crush. The aim of this study was to determine the effectiveness of such treatments to repair the neuronal cytoskeleton evaluating the variation in the number of neurofilaments. For this an experimental design was performed, which involved 30 rats divided into 6 groups: 1 - control healthy; 2 - control injured; 3 - irradiated by LLL 2 J/cm2; 4 - irradiated by LLL 10 J/cm2; 5 - irradiated by US 0.5 W/cm2 and 6 - irradiated by US 1W/cm2. With the exception of group 1 all specimens were anesthetized and underwent right sciatic nerve compression using 40N pressure for 45 seconds. Twenty-four hours after compression irradiation was started by LLL and US according protocol. In our research we found that the increase in the number of neurofilaments was related to the applied dose of LLL and US. The average value of neurofilaments / 0.25 mm2 obtained in each group was: 1 - 128; 2-100; 3-156; 4-140; 5-100; 6-148. We concluded that the application of LLL and therapeutic US increases the number of neurofilaments in rat sciatic nerve undergoing neuropraxia, with LLL being more effective compared to the US. Furthermore we concluded that the effectiveness of therapies to induce regeneration of injured nerve is related to the type of protocol used, demonstrating the need to establish an adequate radiation dose with the purpose of obtaining the best therapeutic response, thus achieving successful treatment.

ABSTRACT

Chondrocytes obtained from stifle joint of New Zealand White rabbits were cultivated. Half of cells were maintained in culture for later implantation and the others frozen during six months to evaluate viability. A circular osteochondral defect was created in the right stifle of other twenty seven rabbits. The control group (CG) received no treatment. The thawed (TH) and fresh (FH) heterologous groups received, respectively, an implant of cultivated thawed or fresh heterologous chondrocytes associated with platelet rich plasma (PRP). The CG group showed greatest pain and lameness compared to the other groups seven days after the implantation. Microscopically, at 45 and 90 days, the TH and FH groups showed filling with cartilaginous tissue containing chondrocytes surrounded by a dense matrix of glycosaminoglycans. In the CG group, healing occurred with vascularized fibrous connective tissue without integration to the subchondral bone. Cryopreserved heterologous chondrocytes were viable for implantation and healing of osteochondral lesions; the association with PRP allows the fixation of cells in the lesion and offers growth factors which accelerates repair with tissue similar to articular hyaline cartilage.

ABSTRACT

The use of medicinal plants and their derivatives for the treatment of diseases is an ancient practice. Currently, there is a growing demand for natural products, not only medicines, but also food and cosmetics. The peppermint (Mentha x piperita L.), in addition to being a medicinal plant, can be used for obtaining flavorings, spices and tea infusions. The drying process is necessary to increase the shelf life of the product and to facilitatg itr transport, handling and storage. For peppermint derivatives of sufficient quality, it is necessary studies on the pre- and post-harvest. The objective of this study is to identify the temperature and air velocity that minimizs the degradation of the color of the leaves with higher oil yield. PepperMint leaves were harvested manually from 7:15 to 8:00 a.m. and dried in trays in a completely randomized design in a 2 x 5 factorial design, being two air velocities (0.3 and 0.5 m.s-1) and five temperatures (30, 40, 50, 60 and 70 °C) with three replications. The dried material was analyzed for color using a colorimeter with the CIELab scale system based on a color within the DIN 6174 standards (1979). The quantification of the essential oil was performed by hydrodistillation in a Clevenger-type apparatus. The analyzes were performed in triplicate. Ws observed that the yield of essential oil of the dry leaves was highertwhan compared to the yield presented by the fresh leaves. This effect was attributed to the fact that the lower water content in the leaves enables the vapor stream generated in the extractor to promots a more effective drag of the volatile oilcompounds stored in the cells; however, a temperature exceeding 50 °C promotes a reduction in yield. The color of the leaves submitted to drying is different from the color of fresh leaves. According to the DIN 6174 standard, the color of the leaves subjected to drying at the temperature up to 40 °C is "easily distinguishable" and, for the leaves submitted to drying at temperatures higher than 50 °C, the differentiation is "too large". We conclude that to get the maximum yield of essential oil and minimal color degradation, drying should be carried out at a temperature of up to 50 °C. The rate of the drying processes in the rangeofrom 0.3 to 0.5 m.s-1 did not affect the parameters evaluated.

ABSTRACT

The knowledge of the anatomical characteristics of the temporomandibular joint (TMJ) articular surfaces is essentital to enable physicians and dentists to recognize the morphological changes that occur in this articulation in patients with temporomandibular disorders (TMD). Several researchers associate the TMD with changes of TMJ articular surfaces. The careful identification of bone changes related to TMJ is critical, since these abnormalities are associated with signs and symptoms of TMD and the knowledge of TMD signs and symptoms is fundamental for correctly diagnosing and for adequate treatment planning. The aim of this study was to evaluate the morphological characteristics of the TMJ articular surfaces in patients with TMD diagnosed according to the Research Diagnostic Criteria for Temporomandibular Disorders (RDC/TMD). In addition, therelationship between increasing age-osteoarthrosis was evaluated. For the sample we selected 19 patients, 17 female and 2 male, referred to the "Unidad de Trastornos Cráneo Cérvico Mandibulares (UCRACEM) - Universidad de Talca, Chile". The imaging assessment was carried out by Cone-Beam Computed Tomography (CBCT). In the imaging analysis of the articular surfaces 11 joints (28.94 percent) showed normal morphology. The bone changes found were: sclerosis, flattening, erosion, osteoarthrosis, osteophytes, subcondral cysts. We found statistically significant difference between increasing age-osteoarthrosis (p=0.00). Considering our results we concluded that bone changes of the TMJ articular surfaces in patients with TMD are very common, with sclerosis as the most frequent finding. It was also possible to conclude that there was a significant association between increasing age-osteoarthrosis.

ABSTRACT

Pycnodysostosis is a rare genetic syndrome characterized by short stature, obtuse mandibular angle, frontal, parietal and occipital bossing, open fontanels and cranial sutures, midfacial hypoplasia, acro-osteolysis of the distal phalanges, increased bone density, absence or hipopneumatization of the paranasal sinuses and normal laboratory studies. We report the case of a 35-year-old Brazilian man that was referred to a private clinic with history of dysmorphic facies for evaluation. The clinical and radiological features exhibited by the patient led to a diagnosis of pycnodysostosis. We describe the morphological features of pycnodysostosis with emphasis on the clinical and radiographic maxillofacial findings comparing the data obtained from our case with a literature review.

ABSTRACT

The aim of this study is to provide morphometric data of bony structures involved in the surgical technique of zygomatic implants, thus providing information to facilitate the planning and implementation of this procedure. Considering the average values obtained in our study, we conclude that the greatest thickness of the zygomatic bone is in the middle of the bone. We suggest that the upper anchorage of zygomatic implants should not extend to the posterior margin of the zygomatic bone, since the bone thickness at this level does not offer suitable conditions for osseointegration of implants.

ABSTRACT

Pain control is one of the most important aspects contributing that the patient has a positive behavior in the dental office. However, this still constitutes a challenge for the dentist when treating anxious patients, because the administration of local anesthesia is the cause of many patients to avoid dental treatment. With the purpose of reducing pain sensation for patients and promote greater comfort for patients and professionals was developed a computerized anesthesia injection method. The aim of this investigation was to compare the conventional local anesthetic technique with the computer-controlled local anesthetic. Participated in this study 30 volunteers patients who had indication for bilateral surgical extraction of lower third molars. The patients, randomly selected, were subjected to each of the two mentioned anesthetic techniques (conventional and computer-controlled). Although the degree of professional satisfaction with the computer-controlled local anesthetic technique has been significant, the most of them reported to prefer the conventional method. There was no significant hemodynamic changes in any of the two methods used in this study. The computer-controlled local anesthetic technique was simple, effective and safe. The patients enrolled in this study responded better to the computer-controlled local anesthetic technique, reporting less pain.

ABSTRACT

Among the techniques used in orthognathic surgery is the Le Fort I osteotomy in which a important surgical step is the disjunction of the spheno-maxillary suture. During this procedure surgical complications, such as neurovascular injuries sometimes occur, mainly due to lack of knowledge of regional anatomy and poor positioning of chisels. The aim of this work is to contribute for this surgical technique to become more predictable, offering morphometric data of the main anatomical landmarks of the pterygopalatine region. The height of the spheno-maxillary suture was on average 13.22mm in men and 12.47mm in women; the width of the spheno-maxillary suture was on average 11.40mm in men and 11.02mm in women, the distance from the point of greatest concavity of zigomaticoalveolar crest until spheno-maxillary suture was on average 25.12mm in men and 23.80mm in women; the distance between the lowest portion of the spheno-maxillary suture and the inferior orbital fissure was on average 31.12mm in men and 29.61mm in women. The values obtained in our study may be used as a parameter in performing the technique of spheno-maxillary suture disjunction, making it more predictable and with less risk of complications.

ABSTRACT

Forty-tree ewes were assigned to four treatments in a randomized block design. The treatments consisted of two flushing diets, composed of soybean meal (SMF) or corn gluten and cottonseed meal (CG+CMF) furnished for 28 days before ovulation, and feeding with flushing diets or a maintenance diet (MD) for 28 days after ovulation: T1 - (SMF) before and after ovulation (n=12); T2 - SMF before ovulation and a MD after (n=11); T3 - CG+CMF before and after ovulation (n=10); T4 - CG+CMF before ovulation and MD after (n=10). The gain in BW and the final BW did not differ (P>0,05) between treatments. The final BC was higher (P>0,05) in ewes in T3 treatment than in those in treatment T2. The urea N concentration was higher (P<0,05) in ewes in treatments T1 and T3 than those in treatments T2 and T4. The pregnancy rate and prolificacy did not differ (P>0,05) between treatments. The flushing diets before ovulation and feeding with this same diet or a maintenance diet after ovulation did not alter the pregnancy rate and prolificacy.

ABSTRACT

The hyperdontia teeth is an anomaly of unknown cause, characterized by increased numbers of teeth. It occurs more frequently in the maxilla, preferably in males, and is more common in permanent dentition. The presence of supernumerary teeth can be responsible for alterations in the permanent dentition, such as the delay in the eruption and dislocations. In addition, the patient may have abnormal occlusion, resorption of adjacent teeth and cyst formation. This article describes in a male patient, 9 years old, the presence of two upper right lateral incisor and one upper left lateral incisor, permanent, partially erupted. Radiological examination revealed a second left upper lateral incisor, unerupted. It should be noted that both, in the region of the 1.2 as in 2.2, the teeth showed normal anatomy and not the conoid appearance commonly observed in supernumerary teeth. In our case, in order to perform orthodontic treatment surgical removal of the teeth noted above was indicated.

ABSTRACT

This work evaluated the effect of freezing on chondrocytes maintained in culture, aiming the establishment of a cell bank for future application as heterologous implant. Chondrocytes extracted from joint cartilage of nine healthy New Zealand White rabbits were cultivated and frozen with the cryoprotector 5 percent dimethylsulfoxide for six months. Phenotypic and scanning electron microscopy analyses were carried out to identify morphological and functional differences between fresh and thawed cells. After enzymatic digestion, a total of 4.8x10(5)cells per rabbit were obtained. Fresh chondrocytes showed a high mitotic rate and abundant matrix was present up to 60 days of culture. Loss of phenotypic stability was notable in the thawed chondrocytes, with a low labeling of proteoglycans and weak immunostaining of type II collagen. The present study showed important loss of chondrocyte viability under the freezing conditions. For future in vivo studies of heterologous implant, these results suggests that a high number of cells should be implanted in the host site in order to achieve an adequate number of viable cells. Furthermore, the chondrocytes should be implanted after two weeks of culture, when the highest viability rate is found.
